My wife and I both used the same vehicle for our businesses. For 'Total kilometres driven in the taxation year', do we both enter the total km's on our returns?

Total kms for my business: ~4000

Total kms for my wife's business: ~6000

What do we each enter for 'Total kilometres driven in the taxation year?'

The total kms for the vehicle(s) for the year will be the same on both.  The kms driven for business will vary between the two of you.
