Yes, you sure can, as long as your wife is considered disabled, and you have a signed T2201 from your physician.
To claim the Home Accessibility Tax Credit, you will go to your "Deductions" tab - "Payment Profile" and you will see the "Home Accessibility Tax Credit" open it and start from there.
