Does Groupon Ads qualify for advertising deduction for small business? They take 50% of your income from each coupon that gets booked

They advertise you online and take 50% of your fee for each customer that books an appointment for reflexology. So, if you had 50 people book at 25.00 a session, they would take 625.00 of the 1250.00 in fees advertised with Groupon.

Yes, this would qualify as an advertising expense.  Ensure that you are claiming the full amount of income (i.e. $1,250 from your example) in your gross revenue and then deduct the 50% cut for Groupon as an Advertising expense.
