Can I determine my tax refund by using my last paycheque in December 2020?

If so, which lines should I use?

Some paystubs have a lot of needed information to include on your return, especially, if they have the year's accumulated amounts. You may be able to estimate a refund but having a completed T4 information slip to use would give you a better result. As per what lines to include the amounts on, you will find our TurboTax software's interview questions easy to understand and follow.
