Is the amount in Box 135 on T4A included in "Medical Expenses"? The total on summary sheet does not equal total of my entered medical expenses + the amount in Box 135.

If you have entered the box 135 amounts from your T4, those amounts transfer to the medical expenses section and do not need to be entered again. 

If you feel that medical expenses have been calculated incorrectly, please contact our phone support team for further assistance.
