On T3 there are two boxes for 52 and 53. Do they both need to be filled in ?

I get an error saying 21 must equal a total of 52 and 53. The numbers are correct. Why the error ?

For the error on the T3 slip not balancing box 52+53 to equal box 21A, trying to enter the sum of 52+53 into box 53 (deleting 52) should fix the problem.
