If you are preparing a coupled return (both your return and your wife's at the same time), you'll just enter both of your income, expenses, credits, etc. as you go through your return.
If you are preparing only your return, when prompted, enter your wife's details such as date of birth, social insurance number, net income, etc.
TurboTax uses your entries to assign credits such as the spouse amount automatically.
To add your children to your return, you'll need to list them as Dependants in the setting up stage of your return.
